The anxiolytic effects of a Valerian extract is based on Valerenic acid

Abstract: BackgroundValerian is commonly used for the treatment of insomnia and anxiety. Valerian extracts allosterically modulate GABA-A receptors and induced an anxiolytic activity. This activity is closely related to valerenic acid. In the present experiments it was investigated whether acetoxy valerenic acid may interfere with the anxiolytic action of valerenic acid.MethodsSituational anxiety was measured using male CD-1 mice in the elevated plus maze test after oral administration of the test substances. In additio… Show more

“…Valerenic acid as the principle constituent was identified as a subunit-specific allosteric modulator of GABA-A receptors, specifically acting as an agonist at β2/3 subunits in male RjHan:WI rat cortical tissues and in Xenopus oocytes tissue investigations, likely to interact with the loreclezole binding pocket (Becker, Felgentreff, Schroder, Meier, & Brattstrom, 2014;Khom et al, 2007Khom et al, , 2016Trauner et al, 2008).…”

“…Valerian, obtained mainly from the roots of Valeriana officinalis L. (Valerianaceae), is a natural agent broadly used as an alternative therapy for sleep disorders and anxiety (Houghton, 1999;Gooneratne, 2008;Becker et al, 2014;Gromball et al, 2014, Gharib et al, 2015. According to US National Library of Medicine of National Institute of Health (NIH), the recommended doses for humans for inability to sleep (insomnia) are 400 to 900 mg of valerian extract up to 2 h before bedtime for as long as 28 days.…”
