The Antiviral Efficacy of Simian Immunodeficiency Virus-Specific CD8 + T Cells Is Unrelated to Epitope Specificity and Is Abrogated by Viral Escape

Abstract: CD8؉ T lymphocytes appear to play a role in controlling human immunodeficiency virus (HIV) replication, yet routine immunological assays do not measure the antiviral efficacy of these cells. Furthermore, it has been suggested that CD8؉ T cells that recognize epitopes derived from proteins expressed early in the viral replication cycle can be highly efficient. We used a functional in vitro assay to assess the abilities of different epitope-specific CD8؉ T-cell lines to control simian immunodeficiency virus (SIV… Show more

“…However, we did not find persistent epitopes to be associated with lower-magnitude CD8 ϩ T-cell responses or lower functional avidities as measured by IFN-␥ ELISPOT assays. Others have also reported that IFN-␥ production (42,58) and the functional avidity (6, 62) of IFN-␥-secreting CTLs may not reflect their antiviral effect (46). Therefore, further studies are required to evaluate the antiviral effect of evolving and persistent epitopes.…”

“…The antiviral activity of AIDS virus-specific CD8 ϩ T cells is well documented in both in vivo (1,4,21) and in vitro (8,24,29) studies. Accordingly, human immunodeficiency virus (HIV) vaccine modalities that focus on engendering antiviral CD8 ϩ T cells are being developed (13,26,28).…”

“…We performed in vitro viral-suppression assays (VSAs) as previously described (24). Briefly, CD8-depleted, phytohemagglutinin-stimulated lymphocytes were infected with SIVmac239 at an MOI of 5 ϫ 10 Ϫ5 and incubated with SIV-specific CD8 ϩ T-cell clones for 7 days.…”
