The Antioxidant Effects of Genistein Are Associated with AMP-Activated Protein Kinase Activation and PTEN Induction in Prostate Cancer Cells

Abstract: Epidemiological evidence suggests a lower incidence of prostate cancer in Asian countries, where soy products are more frequently consumed than in Western countries, indicating that isoflavones from soy have chemopreventive activities in prostate cells. Here, we tested the effects of the soy isoflavone genistein on antioxidant enzymes in DU145 prostate cancer cells. Genistein significantly decreased reactive oxygen species levels and induced the expression of the antioxidant enzymes manganese (Mn) superoxide d… Show more

“…Our study showed that high dose of CDDP caused apoptosis of granulosa cells of ovary at S phase, which was consistent with previous reports that chemotherapeutics have significant role in interfering with cell cycle (Horowitz et al, 2004;Crescenzi et al, 2006;Xu et al, 2007;Hara et al, 2006). Genistein has been shown to have many biological activities, such as anti-cancer, anti-oxidant, antiinflammatory actions and inhibition of tyrosine-specific protein kinases (Akiyama et al, 1987;Rusin et al, 2010;Park et al, 2010;Zhang et al, 2008). GEN has become a popular candidate for drug development because of these features.…”

“…E-mail: haoai358@yahoo.cn. failure caused by chemotherapy and study new technologies and introduce new drugs to prevent and treat the POF.Genistein (GEN) is a phytoestrogen that occurs naturally in the diet and is found in a wide variety of plantderived foods especially in soybeans and soy-based foods (Park et al, 2010). GEN has a well known weak estrogenic effect by binding to estrogen receptors (Kim et al, 1998).…”

“…Genistein also exerts anticancer properties by modulating genes that regulate the cell cycle and apoptosis (Sarkar et al, 2002;Banerjee et al, 2008). As an antioxidant, genistein decreases reactive oxygen species levels, and it also induces the expression of the antioxidant enzymes superoxide dismutase (SOD) and catalase, which are associated with AMP-activated protein kinase (AMPK) and phosphatase and tensin homolog deleted from chromosome 10 (PTEN) pathways (Park et al, 2010). Genistein has been proposed as a natural alternative to estrogen replacement due to its ability to act via estrogen receptor-dependent mechanisms, utilizing the phosphatidylinositol 3-kinase /Akt pathway (Tissier et al, 2007).…”
