The Antinociceptive Actions of Kava Components in Mice

Abstract: 1. The antinociceptive properties of the aqueous extract of the intoxicating beverage kava, and of the lipid soluble extract (kava resin) were tested in mice, by the tail immersion and abdominal constriction methods. Both extracts showed analgesic effects in both tests. 2. Eight purified pyrones from the lipid soluble extract were also tested for activity in the tail immersion test, and kawain, dihydrokawain, methysticin and dihydromethysticin were found to be very effective in producing analgesia. Using the t… Show more

“…When kavalactones were tested individually, they did not exhibit biological activity similar to that found in the whole kava extract. On the other hand, the recombined kavalactones exhibited biological activity found with the entire kava extract (5,13,24).…”

“…Jamieson and Duffield (24) determined that the lipid soluble extract (kava resin) orally administered to male Balb/c mice had a LD 50 of 700 mg/kg and above, with the death due to respiratory failure. The acute toxicity values LD 50 (mg/kg) of the six major kavalactones in mice, dogs, cats, and rabbits were determined and the results are summarized in Table 1 (NLM, 1998).…”

“…Investigations of kava preparations have demonstrated several biological activities. [3][4][5][6] Preparations are used in Europe and North America to manage mild anxiety disorders. 6) Chemical investigations of kava have led to the isolation and characterization of 19 styryl-a-pyrones (kavalactones) which are believed to be responsible for the pharmacological activities.…”
