The Anti-Arthritic Efficacy of Khellin Loaded in Ascorbyl Decanoate Nanovesicles after an Intra-Articular Administration

Abstract: Osteoarthritis is the most widespread joint-affecting disease. The management of persistent pain remains inadequate and demands new therapeutic strategies. In this study, we explored the pain relieving and protective properties of a single intra-articular (i.a.) injection of khellin loaded in nanovesicles (K-Ves) based on ascorbyl decanoate plus phosphatidylcholine in a rat model of osteoarthritis (OA) induced by monosodium iodoacetate (MIA) treatment. The developed nanovesicles (approximately 136 nm) had a na… Show more

“…Quantitative analyses of khellin were carried out using an 1100 High-Performance Liquid Chromatograph (HPLC) equipped with a Diode Array Detector (DAD) (Agilent Technologies Italia Spa; Rome, Italy), and an Eclipse XDB-C18 column with an internal diameter of 3.5 µm and (150 × 4.6) mm dimensions [37]. All the analyses were performed using a gradient analytical method with a flow rate of 0.4 mL/min and a mobile phase composed of (A) formic acid/water (pH 3.2), (B) acetonitrile.…”

“…These figures were similar due to the high affinity of khellin towards the lipophilic environment of the stratum corneum forming momentous depots because of the low value of octanol/water partition coefficient (ca. 3), the melting point (154 – 155 °C) and molecular weight (260.24 Da) 15 , 16 , 17 .…”

“…The in vitro drug diffusion and release kinetics were carried out using Franz cells; the system consists of two chambers, donor and acceptor, separated by a horizontally oriented membrane according to a previous study [17]. Cellulose nitrate filters (Sartorius Stedim Biotech GmbH, Germany), with 0.45 µm pore sizes and 130 µm thickness, were used.…”

“…Neurological function and balance capacity were assessed using a modification of the Longa mothed ( Longa et al, 1989 ) and the balance beam test ( Vanti et al, 2021 ), respectively. The standards of neurological function score and beam balance test are listed in Supplementary Tables S2,S3 .…”
