The Anti‐Alpha‐Actinin Test Completes Anti‐DNA Determination in Systemic Lupus Erythematosus

Abstract: In murine systemic lupus erythematosus (SLE) models, nephritogenic anti-dsDNA IgG has been shown to cross-react with a kidney antigen, alpha-actinin, and to be critical in renal pathogenesis. In humans, studies of anti-alpha-actinin antibodies (Abs) are scarce, and these antibodies remain to be evaluated. We have thus far tested sera from patients with SLE (n = 103), rheumatoid arthritis (RA, n = 93), and primary Sjögren syndrome (pSS, n = 34), and from healthy subjects (n = 160), for the presence of anti-alph… Show more

“…Overall, the mean level of Ab binding to α-actinin (by OD) in the disease subgroups was quite low. Both mean levels and cutoff levels reported here are, however, in agreement with other findings on anti α-actinin Ab binding in humans [45,47,48]. Nonetheless, it may be argued that such low OD values are not meaningful, especially as the potential pathophysiological significance of the presence of α-actinin Abs remains unclear.…”

“…46 The list includes antidsDNA, antihistones, 47-52 and antibodies versus endogenous antigens, such as antiactinin. 10,13,53,54 Actually, definition of specific renal and serum levels of the above antibodies are in progress and will be reported very soon (G.M. Ghiggeri, unpublished data).…”

“…The collagen-like region of C1q protein is believed to play a role in lupus nephritis and autoantibodies to this protein occur in about 30-50% of lupus patients [38]. Anti-alpha-actinin-2 antibodies have been detected in patients with lupus nephritis [40]. It was interesting that these researchers detected anti-alpha-actinin-2 antibodies even before lupus nephritis was present.…”
