Odorant-binding proteins (OBPs) and chemosensory proteins (CSPs) of insects are thought to play roles in olfactory recognition affecting host choice, copulation, reproduction and other behaviors. Previous descriptions of OBPs and CSPs in the whitefly Bemisia tabaci often provided no or incomplete genetic information. In this study, we present a genome-wide and transcriptome-wide investigation of the OBPs and CSPs in B. tabaci MEAM1 (Middle East-Asia Minor1 species). Eight OBP and 19 CSP genes were identified that covered all previous sequences. Phylogenetic analyses showed that the CSP genes had a lineage-specific expansion (BtabBCSP1, BtabBCSP3, BtabBCSP13, BtabBCSP17, BtabBCSP18 and BtabBCSP19). Expression profiling of OBPs and CSPs by transcriptome sequencing and quantitative real-time polymerase chain reaction (qPCR) revealed that expression patterns differed among developmental stages of B. tabaci MEAM1. Five OBP genes and 11 CSP genes significantly differed between males and females; four of the 19 CSP genes were highly expressed in adults, while two were highly expressed in nymphs. The expression profiles of the OBP and CSP genes in different tissues of B. tabaci MEAM1 adults were analyzed by qPCR. Four OBP genes found in B. tabaci MEAM1 were highly expressed in the head. Conversely, only two CSPs were enriched in the head, while the other six CSPs were specifically expressed in other tissues. Our results provide a foundation for future research on OBPs and CSPs in B. tabaci.

Odorant binding proteins (OBPs) are believed to be important for transporting semiochemicals through the aqueous sensillar lymph to the olfactory receptor cells within the insect antennal sensilla. Here, we injected AlinOBP4-siRNA into the conjunctivum between prothorax and mesothorax of the lucerne plant bug, Adelphocoris lineolatus and evaluated the silencing of AlinOBP4 by reverse transcription polymerase chain reaction (RT-PCR) analysis, quantitative real-time PCR (qPCR) test and electroantennogram (EAG) assay. The combination of RT-PCR and qPCR analyses revealed that the levels of messenger RNA transcript were significantly reduced ∼95% in AlinOBP4-siRNA-treated A. lineolatus males and ∼75% in RNAi-treated females within 48 hours. It was found that there are different EAG responses between male and female bugs when the AlinOBP4 gene was silenced by RNAi. The EAGs of A. lineolatus to two plant volatiles, tridecanal and hexyl alcohol, were reduced 9.09% and 79.45% in RNAi-treated males, 62.08% and 62.08% in RNAi-treated females compared to the controls, separately. Antennae of RNAi-treated bugs showed significantly lower electrophysiological responses to four sex pheromone analogs, butyl butanoate, 1-hexyl butyrate, (E)-2-hexenyl butyrate and hexyl hexanoate. The EAG recordings were reduced 35.43%, 35.24%, 39.96% and 78.47% in RNAi-treated males and 64.52%, 18.13%, 36.88% and 49.52% in RNAi-treated females, respectively. The results suggested that AlinOBP4 might play dual-roles in the identification of plant volatiles and sex pheromones. It was suspected that AlinOBP4 may have different functions in odor perception between male and female A. lineolatus.

Background: Anopheles stephensi Listen (1901) is the major malaria vector in the Asia, and recently in some regions of Africa. This species includes three biological forms, namely “type”, “intermediate” and “mysorensis” with varying degree of vector competence for malaria parasites. To recognize these siblings of An. stephensi lab strain, we used the morphological features of eggs and several genetic markers i.e. Obp1 (odorant binding protein), mitochondrial oxidases subunit 1 and 2 (COI and COII), nuclear internal transcribed spacer 2 locus (ITS2). Methods: Eggs were collected from individual mosquito (n = 50) and observed for the number of ridges under stereomicroscope. DNA was extracted from female mosquitoes. After amplifying fragments by using different genetic markers (Obp1, COI, COII and ITS2), the PCR products were purified and sequenced using Sanger Sequence Technology. Phylogenetic analysis was performed after aligning query sequences against the submitted sequences in GenBank using bioinformatics software. Results: The range of ridges number on each egg float was 12-13 that corresponds to the mysorensis form. Sequence analysis for COI, COII and ITS2 demonstrated 100%, 99.46% and 99.29% similarity of our species with other Chinese, Indian and Iranian strains of An. stephensi. All the sequences of Obp1 intron I region matched 100% with the previously submitted sequences for An. stephensi sibling C (mysorensis form) from Iran and Afghanistan.Conclusion: The current study elaborately describes the morphological and molecular details (sequence data) of the ‘mysorensis’ form of An. stephensi that could be helpful in elucidating its classification and also in its diﬀerentiation from other biotypes of the same and other similar anophelines species. Our findings confirmed OBP1 as the only genetic marker that successfully recognized our lab strain (mysorensis form/ sibling C). Thus using OBP1 (alone) may be very phenomenal in similar studies in the future. Future studies should involve the development of appropriate and reliable molecular keys (for sibling species identification) complementary to morphological keys.
