The Anatomical Location Shapes the Immune Infiltrate in Tumors of Same Etiology and Affects Survival

Abstract: The tumor immune microenvironment determines clinical outcome. Whether the original tissue in which a primary tumor develops influences this microenvironment is not well understood. We applied high-dimensional single-cell mass cytometry [Cytometry by Time-Of-Flight (CyTOF)] analysis and functional studies to analyze immune cell populations in human papillomavirus (HPV)-induced primary tumors of the cervix (cervical carcinoma) and oropharynx (oropharyngeal squamous cell carcinoma, OPSCC). Despite the same etiol… Show more

“…Within the adaptive compartment, we found dominant, tumour tissue-specific CD8 + and CD4 + T cell populations that displayed a highly similar activated tissue-resident phenotype. Such CD8 + T cell populations have been described in ovarian cancer,44 45 lung cancer46 and recently in melanoma,47 cervical carcinoma48 and CRC,22 23 and their presence was associated with an improved clinical prognosis. Single-cell RNA-sequencing revealed that CD8 + T cells in colorectal tumours showed a cytotoxic profile, indicative of potential antitumour reactivity.…”

“…Mass cytometry (CyTOF) staining and acquisition was performed as generally described. 13 In short, single-cell suspensions of TC-1 tumors were purified by Percoll gradient to remove aggregates and debris, stained with 37 mouse immune markers, and acquired on a Helios mass cytometer (Fluidigm Sciences). The metal-conjugated antibodies are listed in online supplementary file 1.…”

“…Staining was performed as described in detail elsewhere. 29 All reagents were purchased from Fluidigm Sciences unless stated otherwise. In short, samples were incubated with 1 µM Cell-ID intercalator-103Rh to identify dead cells, followed by blockage of mouse Fc-Receptors using anti-mouse CD16/32 (clone 2.4G2, BD Biosciences).…”
