The Analysis of Neurovascular Remodeling in Entorhino-hippocampal Organotypic Slice Cultures

Abstract: Ischemic brain injury is among the most common and devastating conditions compromising proper brain function and often leads to persisting functional deficits in the affected patients. Despite intensive research efforts, there is still no effective treatment option available that reduces neuronal injury and protects neurons in the ischemic areas from delayed secondary death. Research in this area typically involves the use of elaborate and problematic animal models. Entorhino-hippocampal organotypic slice cult… Show more

“…To study the effects of moderate hypothermia and cold‐inducible RBM3 on sustained ER stress in the brain, we used organotypic hippocampal slice cultures from P3 animals as indicated in Fig . 1 A and as described previously (26). When hippocampal slices were challenged with the ER stress inducer thapsigargin (0.3 μM), the increase of phos‐eIF2α, CHOP mRNA, and CHOP protein levels was prevented in the hypothermic group (Fig.…”

“…Clear cell renal sarcoma often presents with nests of cells separated by fibrovascular septae [12], that we also see in our patient sample, constituting a defined tumor microenvironment. Several methods have been applied to investigate tumors of individuals in an approach to personalize treatment including 2D culture of dissociated tumor cells, 3D culture of spheroid tumors [26], patient derived mouse xenografts [27] and organotypic tumor slice cultures [28]. Of these, the organotypic tumor slice culture is the only method that allows one to study primary culture of an entire tumor piece.…”

“…This study took advantage of the OBSC system to explore mechanisms of pathological angiogenesis that are difficult to study in vivo. OBSCs are a potent, underused tool for exploring vascular phenotypes 23,36,37 with few prior studies seeking to explore this in combination with AD models 32,38 . Interestingly, the increased sprouting angiogenesis we observed in TgCRND8 OBSCs is more pronounced than we find in vivo at a similar age.…”
