The adenosine receptor antagonist CGS15943 reinstates cocaine-seeking behavior and maintains self-administration in baboons

Abstract: The adenosine antagonist CGS15943 reinstated cocaine-seeking and functioned as an intravenous reinforcer. The finding that CGS21680 produced a rightward shift in the CGS15943 reinstatement dose-effect curve supports a role of adenosine mechanisms in the reinstatement of cocaine-seeking behavior.

“…This hypothesis might seem at odds with the results of several reports that indicate that caffeine can reinstate or prevent the extinction of cocaine-seeking behaviour (Worley et al, 1994;Schenk et al, 1996;Self et al, 1996;Kuzmin et al, 1999;Green and Schenk, 2002;Weerts and Griffiths, 2003). However, these effects of caffeine appear to be due to A 1 rather than A 2A receptor blockade (Kuzmin et al, 1999;Green and Schenk, 2002, Solinas et al, 2002, 2005Karcz-Kubicha et al, 2003;Quarta et al, 2004Quarta et al, 2005Antoniou et al, 2005).…”

“…Adenosine may inhibit the reward process via A 2A Rs (Baldo et al 1999 ) . In animal models, A 2A R agonists inhibit cocaine selfadministration, while antagonists reinstate this behavior (Knapp et al 2001 ;Weerts and Grif fi ths 2003 ) . Furthermore, mice lacking the A 2A R exhibit attenuated reward processes (Castane et al 2006 ) .…”

“…51 for review). In the present studies, we focus on the moderate dose of caffeine because our follow-up investigations after caffeine, consists of studies of caffeine and cocaine co-administered and at this moderate (middle) dose, caffeine potentiates cocaineseeking behavior in animals (52).…”
