The addition of simvastatin administration to cold storage solution of explanted whole liver grafts for facing ischemia/reperfusion injury in an area with a low rate of deceased donation: a monocentric randomized controlled double-blinded phase 2 study

Abstract: BackgroundLiver transplantation is the best treatment for end-stage liver disease. The interruption of the blood supply to the donor liver during cold storage damages the liver, affecting how well the liver will function after transplant. The drug Simvastatin may help to protect donor livers against this damage and improve outcomes for transplant recipients. The aim of this study is to evaluate the benefits of treating the donor liver with Simvastatin compared with the standard transplant procedure.Patient and… Show more

“…Liver IRI is a complex pathophysiological process involving multiple factors and is an important cause of poor prognosis and multiple organ failure in patients with complex liver surgery and liver transplantation (Pagano et al, 2018). Our current research showed that liver ischemia for 1 h followed by reperfusion for 6 h led to a significant increase in ALT and AST levels.…”

“…Liver IRI is a complex pathophysiological process involving multiple factors that is associated with a cause of poor prognosis and multiple organ failure in patients after liver transplantation (14). Previous studies demonstrated that the liver IR is characterized by oxidative-stress responses and release of ROS that directly lead to tissue damage and initiates a cascade of deleterious cellular responses leading to in ammation, cell death, and ultimate organ failure (15,16).…”
