The Acute Pancreatic Effect of Alloxan in the Rabbit

Abstract: The origin of the pronounced hypoglycaemic phase during the onset of alloxan diabetes in the rabbit has been investigated.Blood sugar and serum insulin levels were recorded 6, 12 and 24 hr. after alloxan administration and were related to changes in the insulin content of the pancreas and to the rate of insulin release from pancreas slices in vitro at similar time intervals. During the phase of hypoglycaemia, serum insulin levels were elevated, and insulin release from the pancreas slices was markedly increase… Show more

“…Thus, in the untreated or undertreated diabetic patient, hyperlipidemia often occurs. 5) Recent reports demonstrated that oral administration with doses of 400, 600 and 800 mg/kg of the n-butanol fraction of B. forficata reduced the hyperglycemia in alloxan-induced diabetic rats. 10) Pepato et al 11) demonstrated that chronic administration (31 d of treatment) of B. forficata decoction to the diabetic group caused a significant reduction in plasma glucose.…”

“…4) In preliminary work we showed that the crude aqueous extract from B. forficata has a hypoglycemic effect. 5) In this paper we investigate further the effects of oral daily treatment for 7 d with extracts of Bauhinia forficata on glucose and lipid levels of alloxan-diabetic rats.…”

“…Since the early hyperglycemia which follows alloxan has been attributed to an adrenaline response (13), it is reasonable to consider a similar explanation for streptozotocin, although this interpretation has not as yet been tested experimentally. The hypoglycemic phase has been attributed, in the case of alloxan, to the release of insulin from necrotizing pancreatic P-cells, although elevated serum insulin levels have been reported only very recently (14,15). For streptozotocin, the insulin-induced nature of the hypoglycemic phase is fully established by the results shown in Fig.…”

“…In the present paper we report evidence for identical changes in the structure and catalytic properties of FruP2ase in animals treated with triamcinolone or rendered diabetic with alloxan, and parallel changes in the lysosomal proteases that may be responsible for the structural modifi- (16). Approximately 5 days after the injection of alloxan, blood glucose was determined by the glucose oxidase method and animals whose blood glucose levels exceeded 600 mg/100 ml were employed for the experiments described.…”
