The active transport of 5‐hydroxyindol‐3‐ylacetic acid and 3‐methoxy‐4‐hydroxyphenylacetic acid from a recirculatory perfusion system of the cerebral ventricles of the unanaesthetized dog

Abstract: SUMMARY1. An operation on dogs for the implantation ofguide tubes to the lateral ventricle and cisterna magna and a method whereby the ventricular space can be repeatedly perfused in conscious and unrestrained animals are described.2. The characteristics of a recirculatory perfusion system were examined and the bulk formation and absorption of cerebrospinal fluid and the volume of the ventricular space perfused were derived from the concentrations achieved during the infusion of inulin into the system. 3. 5-hy… Show more

“…Animal methodology The animals used in these experiments were adult male beagles, 10-14 kg which had had guide tubes directed towards the lateral ventricles implanted in the skull (Ashcroft, Dow & Moir, 1968b) at least two weeks before any subsequent experimental procedure.…”

“…From the rate of infusion of exogenous 5-HIAA and the equilibrium concentrations of 5-HIAA in the perfusion system before and during this infusion, the rate at which exogenous 5-HIAA was removed from the c.s.f. system was calculated (Ashcroft et al, 1968b). It was assumed that this was also the rate at which endogenous 5-HIAA was removed from the c.s.f.…”

“…Four male Beagle dogs with indwelling ventricular cannulae (Ashcroft, Dow & Moir, 1968) had 0.5 ml of CSF withdrawn on 3 occasions, at 0, 30 and 60 min before an intravenous loading dose of equal volumes of 0.9% w/v NaCl solution (saline), or D or Ltryptophan (50mg/kg made up in saline at a concentration of 20 mg/ml). This dose was chosen as that producing a change in 5-HT synthesis when cisternal CSF was used as an index of cerebral turnover (Eccleston, Ashcroft, Moir, Parker-Rhodes, Lutz & O'Mahoney, 1968).…”

“…It is localized in the region of the fourth ventricle (8). In CSF, there is a gradient in the homovanillic acid concentration: in man, the concentration falls from 466 £/1 in the ventricular CSF to 185 in the cisternal and to 53 /ig/1 in the lumbar CSF (9).…”
