The accessibility of protein-bound dinitrophenyl groups to univalent fragments of anti-dinitrophenyl antibody

Abstract: A series of N-(N-dinitrophenylaminoalkyl)maleimides were sythesized with alkyl-chain lengths of two, four and six carbon atoms. When these compounds reacted with the thiol group of mercaptalbumin, the tryptophan fluorescence of the protein was quenched. This change in fluorescence was used to determine the rate of reaction of the Dnp (dinitrophenyl)-maleimides with mercaptalbumin. The second-order rate constants were similar to those observed in reactions between low-molecular-weight thiol compounds and maleim… Show more

“…Dicyclohexylcarbodi-imide and picric acid were from BDH Chemicals, Poole, Dorset, U.K. 1-Hydroxybenzotriazole hydrate was from Aldrich Chemical Co., Gillingham, Dorset, U.K. 2-Naphthylamine hydrochloride and dithiothreitol were from Sigma Chemical Co. Ltd., Poole, Dorset, U.K. Other reagents were obtained commercially and were of the highest purity available. Dimethylformamide was further purified by the method of Knight & Green (1976).…”

“…Anti-Dnp antibody was produced in rabbits (Eisen, 1964) and purified as described by Eisen et al (1967). A univalent (Fab) fragment preparation (Knight & Green, 1976) gave only one protein band on slab-gel electrophoresis (Davies et al, 1978), corresponding to mol.wt. 45000.…”

“…Values of the apparent dissociation constant, Kapp.' were calculated as described by Knight & Green (1976 0.1 x 10-9M at 0.04,uM-antibody, indicating that most of the antibody species in the preparation bound the hapten with Kapp < 10-10M (cf. Knight & Green, 1976).…”

“…We have described previously (Knight & Green, 1976) a new technique for probing the environment of the amino acid residues in proteins that can be labelled with Dnp groups. Fluoresence-quenching titrations showed that Dnp groups attached by aminoalkylsuccinimide chains to the thiol group of bovine mercaptalbumin were fully available to univalent fragments of high-affinity antibody to Dnp, even when the alkyl chain was short.…”
