“…Synthetic BLP, palmitoyl-3-cysteine-serine-lysine-4 (Pam 3 CSK 4 ), was obtained from EMC Microcollections (Tübingen, Germany). Streptococcal protein I/II was prepared as previously described (20). Potential endotoxins in the protein I/II preparation were removed using polymyxin B-agarose (Pierce, Rockford, IL) according to the manufacturer's recommendations.…”

“…Purification of Protein I/II-Recombinant protein I/II of S. mutans OMZ 175 was purified from pHBsr-1-transformed Escherichia coli cell extract by gel filtration and immunoaffinity chromatography as previously described (31). The purity of the protein was checked by SDS-PAGE after staining with Coomassie Blue.…”

“…Recombinant protein I/IIf and protein PD 26 from Streptococcus mutans OMZ 175 were puri®ed from pHBsr-1or pSR D 26 transformed E. coli cell extract, respectively (Ogier et al, 1989), by ammonium sulphate precipitation, followed by fractionation on an anti-protein I/IIf IgG Sepharose column and further puri®cation by anion exchange chromatography (Chatenay-Rivauday et al, 1998). Protein I/IIf migrated as a single band with an apparent molecular mass of 195 kDa, as checked by SDS±PAGE after staining with Coomassie blue.…”

“…Moreover, we and others have demonstrated the capacity of protein I/IIf to bind to extracellular matrix proteins, such as ®bronectin, collagen I and laminin (Love et al, 1997;Sciotti et al, 1997), and to epithelial, endothelial and monocytic cells (Soell et al, 1994;Vernier et al, 1996). We demonstrated recently that the speci®c binding of protein I/IIf from Streptococcus mutans serotype f to monocytic THP-1 cells occurs via lectin interactions speci®c for N-acetyl neuraminic acid (NANA) and fucose and is controlled by the extended V-region (Chatenay-Rivauday et al, 1998). We also showed that (i) the binding of members of the oral viridans streptococci to cells induces the release of pro-in¯ammatory cytokines by epithelial and endothelial cells (Vernier et al, 1996); (ii) protein I/IIf belongs to the recently de®ned class of modulins and exerts immunomodulatory effects on human cells (Soell et al, 1994;Vernier et al, 1996).…”
