The 5-HT1A agonist 8-OH-DPAT increases consumption of palatable wet mash and liquid diets in the rat

Abstract: The 5-HT1A agonist 8-OH-DPAT, at a dose of 30 micrograms/kg, enhanced the consumption of sweetened wet mash and sweetened milk in non-deprived rats. In partially satiated rats, sensitivity to the hyperphagic effect of 8-OH-DPAT on wet mash intake was substantially increased, so that the minimally effective dose was reduced to 3 micrograms/kg. Similarly, 8-OH-DPAT was more efficacious in increasing milk intake in satiated rats. Thus, 30 and 40 micrograms/kg 8-OH-DPAT produced a 4-fold increase of milk intake in… Show more

“…Previously, we have observed similar increases in the consumption of food pellets after comparable doses of 8-OH-DPAT and gepirone (Dourish et al 1985a;Gilbert and Dourish 1987). Similar- (Dourish et al 1988). However, experiments which have examined the effects of 8-OH-DPAT on the consumption of sweetened liquids have produced conflicting results.…”

“…This observation indicates that 8-OH-DPAT does not elicit non-specific chewing. These data, together with the recent reports that 8-OH-DPAT increases milk intake (Dourish et al 1988) and sham feeding , provide compelling evidence that hyperphagia induced by the drug is due to a specific action on brain 5-HT mechanisms which control feeding.…”

“…These studies in humans confirm reports in animals that m-CPP decreases food intake, including carbohydrates, protein, and fat in a mixed testmeal. [Neuropsychopharmacology 11:63-71, 1994J man generally leads to increased satiety (Blundell 1984(Blundell , 1986, with the exception of 5-HT IA receptor activation that induces feeding in animals in specific paradigms (Dourish et al 1985(Dourish et al , 1986(Dourish et al , 1988.
