“…2), suggesting a regulatory role, such as the control of transcription by RNA polymerase III (Nederby-Nielsen et al 1993;Pendás et al 1994;Suzuki et al 1996). Moreover, as in the NTSs of other fish species (Felgenhauser et al 1990;Pendás et al 1994;Sajdak et al 1998;Martins et al , 2001, a TATA-like sequence was observed upstream of the 5S rRNA gene in Brycon, at the -19 to -27 position (Fig. 2).…”

“…Upstream (5Ј) control regions have been defined for the somatic 5S of X. laevis (Oei and Pieler 1990), the loach [Misgurnus fossilis (Felgenhauer et al 1990)], and the silkworm [Bombyx mori (Morton and Sprague 1984)]. Study of deletion mutants has shown that upstream control elements are required for wild-type expression.…”

“…Note 1: Testing of deletion mutants in transcription assays showed reduced levels of transcription when 5Ј flanking regions were shorter than these (Oei and Pieler 1990;Felgenhauer et al 1990;Morton and Sprague 1984). (Pieler et al 1987).…”

“…In various vertebrate species, a conserved external promoter sequence, a D-box in mammals, is known to be present in the NTS upstream of the 5S rRNA gene (Hart and Folk 1982;Nielsen et al 1993;Hallenberg et al 1994;Jensen and Frederiksen 2000;Frederiksen and Andersen 2003;Fujiwara et al 2006), besides an internal control region that functions as a promoter for transcription of the gene by RNA polymerase III (Hallenberg et al 1994). Also, a similar cis-acting element influencing the transcriptional activity is located in the region between position -18 and the transcription start point in the 5S rRNA gene of loach oocyte (Felgenhauer et al 1990). Taking altogether, the conserved NTS portion of the bitterling 5S rDNA may function as an external promoter such as a mammalian D-box.…”
