The 4-hydroxybenzoate/4-aminophenazone chromogenic system used in the enzymic determination of serum cholesterol.

Abstract: A single reagent, containing cholesterol oxidase, cholesterol esterase, peroxidase, 4-hydroxybenzoate, and 4-aminophenazone, is used in determining serum cholesterol. Analysis time is 15 min, and the standard curve is linear to 6.0 g/liter. Analytical recovery of cholesterol was 100.1 +/- 0.4%. Within-run precision (CV) was less than or equal to 1.4 1.4%, between-run less than or equal to 4.8%. Comparison with results by a Liebermann Burchard method [Clin. Chim. Acta 5, 637 (1960)] gave a linear regression of … Show more

“…Serum glucose concentration was measured by enzymatic method (glucose oxidase p‐aminophenazone) as used by Asadi, Hallajian, Asadian, Shahriari and Pourkabir () with an automatic biochemical analyser (BioTek, Winooski, VT, USA) in our laboratory (Institute of Hydrobiology, the Chinese Academy of Sciences). Serum triglyceride concentration was assayed using glycerol phosphate oxidase/peroxidase‐amido pyrine (GPO‐PAP) method (Fossati & Prencipe ) and total cholesterol concentration was assayed using oxidase⁄peroxidase‐amido pyrine (CHO‐PAP) method (Meiattini, Prencipe, Bardelli, Giannini & Tarli ). All reagents were prepared by Nanjing Jiancheng Bioengineering Institute, Nanjing, China.…”

“…11) exhibited significant anti-inflammatory activities in different degree both in vivo and in vitro ( Table 10). The SAR study indicated that the presence of the 3-benzo[d] [1,3]dioxole ring system and the N-containing heterocyclic amine attached to the methyl group at 4-position of the antipyrine structure was very essential for activity as in case of compounds 134 and 135.…”

“…Nowadays, multiple FDA approved drugs containing pyrazolone nucleus have been explored (Fig. 2), for instances, edaravone (2) has been used as free radical scavenger for the treatment of amyotrophic lateral sclerosis (ALS) [2], aminophenazone (3) with antipyretic and anti-inflammatory activities has been used in breath tests to measure the cytochrome P-450 metabolic activity in liver function evaluations [3], eltrombopag (4) has been used for the treatment of low blood platelet counts in adults with idiopathic chronic immune thrombocytopenia [4], dichloralphenazone (5) has been used for the relief of tension and vascular headaches [5], metamizole (6) has been used for perioperative pain, cancer pain, acute injury and other forms of pain and is considered as the strongest antipyretic [6]. Several investigational small molecules containing pyrazolone have been regarded as drug candidates including sulfamazone (7) [7], propyphenazone (8) [8] and nifenazone (9) [9].…”

“…[25] The levels of TC, TAG and HDL-C were estimated in plasma colorimetrically using commercial kits. [26][27][28] Low-density lipoprotein cholesterol (LDL-C) levels were calculated as previously described. [29] The atherogenic index was calculated using the formula: atherogenic index = LDL-C/ HDL-C. [30] Thoracic aorta reduced glutathione (GSH) was assessed spectrophotometrically using Ellman's reagent, according to the modified method of Ahmed et al [31] Lipid peroxidation content in thoracic aorta was determined by thiobarbituric acid reaction with lipid peroxide end-product malondialdehyde (MDA).…”
