Th1 and Th2 cytokine profiles in recurrent aborters with successful pregnancy and with subsequent abortions

Abstract: We conclude that abortion-prone women who proceed to have successful pregnancy are more Th2-biased than abortion-prone women who abort, and that recurrent aborters who undergo spontaneous abortion have a stronger Th1 bias than aborters who have normal pregnancy.

“…Several studies have shown an elevated production of Th2-like cytokines and a decrease in Th1-like cytokines during normal pregnancies compared to pathological pregnancies (Marzi et al 1996;Raghupathy et al 2000;Makhseed et al 2001). The distinct Th2-bias during normal pregnancies showed in these studies could be enhanced as a consequence of a control group including both non-allergic and allergic individuals.…”

“…Furthermore, stimulation with paternal leukocytes increases the secretion of IL-4 by maternal PBMC during normal pregnancy (Ekerfelt et al 1997). Previous studies have reported a high secretion of Th2-like cytokines at the time of delivery of normal pregnancies and increased Th1-like cytokines at the time of a spontaneous abortion (Marzi et al 1996;Makhseed et al 2001). Taken together, a local as well as a systemic Th2-deviation during pregnancy may be necessary for a successful pregnancy and the ability to easily direct immune responses to a Th2-polarized profile may be an evolutionary benefit.…”

“…Several similar studies indicate increased Th1-like and decreased Th2-like responses among women with pregnancy losses, whereas women with successful pregnancies tend to have an increased Th2-like response Makhseed et al, 2000;Makhseed et al, 2001;Kwak-Kim et al, 2003). An immune deviation towards Th2 has also been found locally at the maternal-fetal interface in healthy pregnant women , while a decreased local production of Th2 cytokines has been demonstrated in women with unexplained recurrent spontaneous abortions (Piccinni et al, 1998).…”

“…Makhseed et al (13) found significantly higher levels of the type 1 and lower levels of the type 2 cytokines in supernatants of activated peripheral lymphocytes from women undergoing preterm delivery, than in those from healthy pregnant women. Furthermore, abortion-prone women who proceeded to have successful pregnancy were more Th2 biased than abortionprone women who aborted (14).…”
