TGF-β signalling defect is linked to low CD39 expression on regulatory T cells and methotrexate resistance in rheumatoid arthritis

Abstract: Rheumatoid arthritis (RA) is an autoimmune arthropathy characterized by chronic articular inflammation. Methotrexate (MTX) remains the first-line therapy for RA and its anti-inflammatory effect is associated with the maintenance of high levels of extracellular adenosine (ADO). Nonetheless, up to 40% of RA patients are resistant to MTX treatment and this is linked to a reduction of CD39 expression, an ectoenzyme involved in the generation of extracellular ADO by ATP metabolism, on circulating regulatory T cells… Show more

“…We did detect a moderate increase in CD73 and the expression of both CD39 and CD73 can be increased by TGF-β [70,71]. Though CD73 is expressed intracellularly in humans, surface expression can be induced upon activation with high-dose IL-2 therapy [72].…”

“…Consistent with an important role for TGF-β in regulating CD39 expression, carriers of at least one mutant allele for a TGFBR2 SNP (rs1431131) showed a significant reduction of CD39 mRNA levels in CD4 + T cells. 102 Extracellular ATP and adenosine signaling further regulate Treg survival and function. Notably, P2×7 is highly expressed in Tregs.…”
