Visible wavelength surface-enhanced Raman spectroscopy (SERS) has been observed from bovine serum albumin (BSA) using In-InP nanopillars synthesised by Ar gas cluster ion beam sputtering of InP wafers. InP provides a high local refractive index for plasmonic In structures, which increases the wavelength of the In surface plasmon resonance. The Raman scattering signal was determined to be up to 285 times higher for BSA deposited onto In-InP nanopillars when compared with Si wafer substrates. These substrates demonstrate the label-free detection of biomolecules by visible wavelength SERS, without the use of noble metal particles. Surface-enhanced Raman spectroscopy (SERS) utilises the resonant excitation of localised surface plasmons to boost the scattering efficiency from samples containing few or even single molecules 1-3. The technique's usefulness for non-destructive and label-free characterisation has resulted in a large body of research since its discovery in 1974 3,4. Gold and silver nanoparticles have historically been the most often employed SERS substrates due to their strong visible wavelength localised surface plasmon resonance (LSPR) 5,6. The deposit patterns derived from droplet evaporation allow current development of medical tests and new strategies for diagnostic in patients. For such purpose, the development and implementation of algorithms capable of characterizing and differentiating deposits are crucial elements. We report the study of deposit patterns formed by the droplet evaporation of binary mixtures of proteins containing NaCl. Optical microscopy reveals aggregates such as tip arrow-shaped, dendritic and semi-rosette patterns, needle-like and scalloped lines structures, as well as star-like and prism-shaped salt crystals. We use the first-order statistics (FOS) and gray level co-occurrence matrix (GLCM) to characterize the complex texture of deposit patterns. Three significant findings arise from this analysis: first, the FOS and GLCM parameters structurally characterize protein deposits. Secondly, they conform to simple exponential laws that change as a function of the NaCl concentration. Finally, the parameters are capable of revealing the different structural changes that occur during the droplet evaporation.

The relationships between the structural and aggregational state of bovine serum albumin (BSA) and the specific length and total number of zigzag pattern segments of the film textures formed upon drying biopolymer solutions with aluminum and iron chlorides have been shown. To obtain films, saline solutions of BSA were dried in a glass cuvette under thermostatically controlled conditions. It is shown that the formation of zigzag structures is sensitive to the influence of aluminum chlorides Al3+ and iron chlorides Fe3+ and depend on the concentration of AlCl3 and FeCl3. This may be due to a change in the charge and size of BSA particles and due to a change in conformation or a violation of the structure of BSA. These factors, in turn, affect the hydration of the solution components and the structural state of free water in solution, which presumably also affects the formation of zigzag structures. It is established that the analysis of the specific length and the number of segments of zigzag patterns makes it possible to evaluate changes in the state of biopolymers in the initial solution during structural changes and aggregation.
