Tetramethylpyrazine reverses intracerebroventricular streptozotocin-induced memory deficits by inhibiting GSK-3β

Abstract: Brain dysfunction, especially cognitive impairment, is one of the main complications in Alzheimer's disease (AD), which threatens the health of 46.8 million people worldwide. At present, the pathogenesis of cognitive dysfunction is only partially understood, and effective therapies for memory loss in AD remain elusive. Tetramethylpyrazine (TMP) is one of the major bioactive compounds purified from Chuanxiong, a Chinese herb used for the treatment of neurovascular and cardiovascular diseases. The neuroprotectiv… Show more

“…42 A close relationship exists between impairment in behavioral function and disruption of neurotransmitters homeostasis in hippocampus in AD as well as in experimental animals. 43,44 Linked with this, these alterations in hippocampal neurochemistry following ICV-STZ infusion in rats in the present study. On the other hand, significant alterations in hippocampal neurochemistry including deficits in monoamines (NE, DA and 5HT) and disturbed balance of GABA and Glutamate have also been reported to occur in AD 45,46 as well as following ICV-STZ infusion in rats.…”

“…To investigate the basis of compositions against AD, we searched the previous literature for these 9 ingredients and surprisingly found that 6 of them were confirmed to possess anti-AD effects (Table 2 ). For instance, tetramethylpyrazine (TMP), one of the major bioactive compounds purified from CX, shows great potential against AD, which could restore the function of cholinergic neurons and protect against memory loss in AD rat model [ 65 ]. Besides, ligustilide was reported to have neuroprotective effect via inducting α-Secretase processing of both APP and Klotho in AD mouse [ 66 ].…”

“…For example, it is stated that the activation of GSK-3β is closely related to aluminum-induced long-term potentiation injury in rats ( 8 ). Others have shown that tetramethylpyrazine can protect the memory loss of AD patients by inhibiting GSK-3β activity ( 9 ). In recent years, studies have also found that there is a certain regulatory relationship between GSK-3β and miR.…”
