Tetramethylpyrazine Inhibits Activation of Hepatic Stellate Cells through Hedgehog Signaling Pathways In Vitro

Abstract: Background and Aim. Tetramethylpyrazine (TMP), a major alkaloid isolated from Ligusticum chuanxiong, has been reported in hepatic fibrosis models. However, the action mechanism remains unclear. In the present study, effects of tetramethylpyrazine (TMP) against hepatic stellate cell (HSC) activation as well as the possible mechanisms were evaluated. Methods. Western blot assay was used to detect TMP effects on protein expression of Smo, Patched, Hhip, and Gli and to investigate the effects of TMP on Cyclin D1, … Show more

“…10,13 Therefore, chemical inhibitors of hedgehog signaling are promising treatments for liver fibrosis. [14][15][16][17] The inhibitor cyclopamine (Cyc) has been shown to restrain HSC activation and viability both in vitro and in vivo. 12 Another promising anti-hepatic fibrosis agent is curcumin (Cur), a major curcuminoid from turmeric.…”

“…The regulation of the Hh signaling pathway by TMP has also been investigated in numerous studies. For example, TMP was demonstrated to effectively inhibit the Hh signaling pathway in hepatic fibrosis (24), has also been reported to be activated in cervical carcinoma cell lines (25). However, the role of TMP in CC, and the associated underlying mechanisms, remain unclear.…”

“…44 In addition, CXVO exerts insecticidal action, 15 further studies indicating that Z-LIG exhibited remarkable larvicidal activity against S. litura larvae. 61 Moreover, Hu et al 62 discovered that TMP significantly inhibited the expression of cyclin D1, cyclin E1, and cyclin-dependent kinase CDK2, inhibiting the proliferation of hepatic stellate cell (HSC) and treating hepatic fibrosis via the Hh signaling pathway. A recent study showed that CX extract-polylactic acid sustained-release microspheres significantly enhanced both the duration and hepatoprotective efficacy.…”
