Modifications of exposure time, substrate, and formulation were made to the Food and Agriculture Organization Larval Packet Test (LPT) to determine a combination suitable for measuring the susceptibility of Boophilus microplus (Canestrini) to amitraz. Exposure time influenced the slope of the dose-response when paper was used as a substrate for amitraz. However, time did not influence the dose-response slope when nylon fabric was used as an amitraz substrate. Formulated amitraz produced results with less deviation from the log-probit model than technical amitraz. The combination of formulated amitraz and nylon fabric as a substrate for amitraz produced results that best fit the log-probit model. The modified FAO procedure (formulated amitraz/nylon substrate combination) was used to assay a Brazilian strain of B. microplus and a Panamanian strain of Rhipicephalus sanguineus (Latreille). Resistance ratios (95% CI) of 26.3 (25.7-26.9) and 7.3 (5.5-9.9) were calculated for the B. microplus and R. sanguineus strains, respectively. A discriminating dose of 0.03% amitraz was determined for B. microplus. This technique will help to locate amitraz resistant tick populations, provide data for improved control practices, and aid in the discovery of resistance mechanisms through synergist studies and verification of molecular techniques.

In addition to pesticidal activity, the capacity of natural compounds to inhibit the reproduction of parasites emerge as an important alternative tick control method. In this context, Satureja spp. stand out due to their recognized pesticidal properties. Among parasites of veterinary importance, the cattle tick, Rhipicephalus microplus, is responsible for great economic losses in livestock and transmission of relevant pathogens. Thus, the aim of this study was to evaluate the effects of different concentrations of the essential oil of Satureja montana L. on the ovary morphology of R. microplus engorged females through histological and histochemical techniques. The most remarkable morphological changes found were: cytoplasmic vacuolation of germ cells, irregular and thicker chorion, irregular oocyte shape, ring‐shaped nucleolus, decrease in protein and carbohydrate content in oocytes, in addition to cellular changes in the oviduct and pedicel. All morphological changes were assessed using a semiquantitative method already established in the literature. Ticks exposed to 5.0 μl/ml of essential oil showed the most significant changes when compared to control groups. Thus, the essential oil of S. montana L. damaged the reproductive system of R. microplus, which may impair ticks' offspring production and promote a long‐term control of this species. Highlights The essential oil of Satureja montana L. affects the ovary morphology of the cattle tick. The main morphological alterations found were cytoplasmic vacuolation, irregular and thicker chorion and irregular oocyte shape. These alterations may impair the development of eggs.

Parasitism constitutes one of the major problems that affect the livestock. Commercial acaricides can propitiate drug resistance and residues in the environment and animals. This study aimed to evaluate the control of cattle ticks by using the isotherapics. These drugs were prepared using Rhipicephalus (Boophilus) microplus. The dilution and agitations were performed according to the homeopathic pharmacopoeia. By the immersion technique of adults, 50 teleogines were distributed into five treatments: 12cH and 32cH isotherapic, negative control, positive (ivermectin 1%) and vehicle control. The results were evaluated by the rate of mortality, egg production, hatchability rate, estimated reproduction, and the efficacy of the products. In addition, 16 Jersey females cows were subjected to 15mL pour on, of the following treatments: 12cH and 32cH isotherapic and vehicle control. The count of ticks was performed by the simple method on days 0, 15, 21, 42, 63, 84 and 105, post treatments. It was used the Wilcoxon test and Bonferroni method. Both isotherapics treatments showed efficacy at in vitro tests. The 12cH medication presented better results: 10% of mortality rate on the first day of life, maintaining 20% from the second to the 13th day. Additionally, it was observed 20% of egg production, hatching rate of 0.0038% and 99.39% of product effectiveness. However, the in vivo tests showed no satisfactory results for these evaluated conditions, although it was verified a tendency to decreasing the infestation.
