Tertiary Lymphoid Structures Associate with Tumour Stage in Urothelial Bladder Cancer

Abstract: Abstract. Background: Urothelial bladder cancer (UBC) is a highly prevalent disease in North America, however its optimal management remains elusive. The contribution of B cell associated responses is poorly understood in bladder cancer. Lymphoid neogenesis is a hallmark of an active immune response at tumor sites that sometimes leads to formation of tertiary lymphoid structures (TLS) that resemble germinal centers formed in secondary lymphoid organs. Objective: This study was conducted with an aim to investiga… Show more

“…For example, the abundance of TLSs increases in advanced stages (II-IV) compared with stage I gastric cancer (171 patients analysed) 33 . A pilot study (28 patients) indicated that TLSs were more common in high-grade muscle-invasive bladder cancer than in the non-muscle-invasive cancers 91 . In breast cancer, high-grade tumours were found to be twice more prone to exhibit TLSs than low-grade tumours, and the most aggressive tumour grade, grade 3, is an independent predictor for TLS formation in patients with human epidermal growth factor receptor 2 (HER2; also known as ERBB2) − tumours 66,92,93 .…”

“…In addition to lymphoplasmacytic and eosinophilic inflammation, about 15% of all canine UCs presented with formations of one or several lymphoid follicles, typically located in peritumoural tissue. The presence of tertiary lymphoid structures (TLSs) has been reported to occur in bladder and other types of cancer (Koti et al, 2017). Studies of the prognostic significance of TLSs in bladder cancer are rare.…”

“…Studies of the prognostic significance of TLSs in bladder cancer are rare. Koti et al (2017) reported a poor prognostic significance of TLSs, which was seen more commonly in high-grade muscle invasive, compared with non-muscle invasive, bladder cancer. In the current series, no association between the presence and extent of TATE or TLSs, and muscle invasive growth, age, sex, neutering status or dog breed was identified.…”

“…22,23 Indeed, our previous report established an increase in TLS density and maturation with advanced stages of bladder cancer. 24 The average age of diagnosis for BC is ~73 years. Biological aging is accompanied by a decline in lymphopoiesis and depletion of naïve B cells from peripheral pools, with a simultaneous expansion of subpopulations such as atypical B cells (ABCs; also known as age-associated B cells).…”
