TERT promoter regulating melittin expression induces apoptosis and G0/G1 cell cycle arrest in esophageal carcinoma cells

Abstract: Esophageal squamous cell carcinoma accounts for a large proportion of cancer-associated mortalities in both men and women. Melittin is the major active component of bee venom, which has been reported to possess anti-inflammatory, antibacterial and anti-cancer properties. The aim of the present study was to construct a tumor targeted recombinant plasmid [pc-telomerase reverse transcriptase (TERT)-melittin] containing a human TERT promoter followed by a melittin coding sequence and to explore the effects of this… Show more

“…This antiproliferative effect on colon cell agrees with literature [ 32 , 48 ]. The low tumor cell viability was also demonstrated on gastric [ 37 ], lung cancer [ 49 ], esophagus [ 34 ], lymphoma [ 29 ], leukemia [ 27 ], ovarian cancer [ 28 ], breast cancer [ 48 , 50 ], cervical cancer [ 43 ], skin cancers [ 20 , 31 ]. Accordantly, the peptide effects on CRC cells have already been demonstrated [ 38 - 40 , 51 ].…”

“…To obtain kk-64 DNA sequence, the KK-64 amino sequence was reverse-translated into DNA sequence using optimized codons for human and synthesized by GENEWIZ (Suzhou, Jiangsu, China). The pcTERT plasmid was constructed as described previously [ 32 ]. kk-64 DNA fragment was digested with HindIII and XbaI and subsequently sub-cloned into pcTERT to generate pcTERT-kk-64 plasmid.…”
