Teratogenic effects of suramin on the chick embryo

Abstract: Suramin, a polysulfonated naphthylamine, has been used for the chemotherapy of trypanosomiasis and onchocerciasis since about the 1920s. Currently, it is also being tested as an anticancer agent. It is hoped that suramin might stop the progression of some kinds of cancer since it has been found to inhibit the proliferation and migration of cells and the formation of new blood vessels. These processes are not only essential for the development and progression of cancer, but also for normal embryonic development… Show more

“…(46)(47)(48)(49)(50)(51) Several prescription drugs, including calcium valproate, acetazolamide, suramin and flubendazole, have also been found to cause body-wallrelated abnormalities in rodents and avians. (52)(53)(54)(55) Given that these substances cause embryotoxicity in rodents and/or chickens, the same birth defects have been inferred in pregnant women. Clearly, however, rodent, avian and human embryos may not be affected in equivalent ways by the same environmental agent.…”

“…Data on experimentallyinduced holoprosencephaly in the golden hamster even suggest that alterations in selected proteins in the prechordal mesoderm and adjacent floor plate, impair the adjacent neural plate and neural crest and, thus, induce these anomalies (Coventry et al 1998). Similarly, the anticancer agent suramin was found to cause midfacial defects in chicken (Männer et al 2003).…”

“…Secondly, among these suramin-treated embryos the open defects occurred with an incidence of 8%, which corresponded to the incidences of cloacal exstrophy found in previous studies on suramin-treated chicks on the 8th incubation day (4-7%; Ma¨nner and Kluth 2003;Ma¨nner et al 2003). Thirdly, the body wall defects open into the embryonic cloaca and allantois (Fig.…”

“…first, these open defects in the infra-umbilical body wall occurred in a small proportion of young chick embryos that had been treated with the drug suramin according to established protocols, which had been proven to induce cloacal exstrophy in chicks (Ma¨nner and Kluth 2003;Ma¨nner et al 2003). Secondly, among these suramin-treated embryos the open defects occurred with an incidence of 8%, which corresponded to the incidences of cloacal exstrophy found in previous studies on suramin-treated chicks on the 8th incubation day (4-7%; Ma¨nner and Kluth 2003;Ma¨nner et al 2003).…”
