Teneurins are type II transmembrane proteins that play important roles in pattern formation in Drosophila, axon fasciculation and organogenesis in Caenorhabidits elegans, and neuronal pathfinding in the visual system of the mouse. There is evidence that a peptide derived from a proteolytic event near the C-terminus of teneurins leads to formation of an active neuropeptide, while processing at and near the transmembrane domain leads to shedding of the extracellular domain into the extracellular matrix and the generation of an intracellular fragment that is transported to the nucleus. In vertebrates there are four teneurins. Here, we have studied the expression of teneurin-4 in the chicken embryo. An antiserum against part of the intracellular domain of teneurin-4 recognizes several low molecular weight bands on immunoblots of embryonic chicken brain homogenates, indicating that teneurin-4 is likely to be processed at one or more predicted proteolytic cleavage sites. Antisera against the EGF-like repeats of the extracellular domain label some mesenchyme in the early embryo, and near basement membranes this labeling partially overlaps with anti-laminin (gamma 1 chain) immunostaining. At embryonic day 7, anti-teneurin-4 labels bundles of axons in the nasal, but not temporal retina. Later in development, retinal expression switches so that teneurin-4 is found in the temporal, but not nasal, ganglion cell layer. Teneurin-4 immunolocalization was also compared with other teneurins in the developing limb, where each teneurin is expressed in distinctive regions. These patterns of expression suggest roles for teneurin-4 in patterning and neuronal pathfinding in the avian embryo.

The formation of visual circuitry is a multistep process that involves cell–cell interactions based on a range of molecular mechanisms. The correct implementation of individual events, including axon outgrowth and guidance, the formation of the topographic map, or the synaptic targeting of specific cellular subtypes, are prerequisites for a fully functional visual system that is able to appropriately process the information captured by the eyes. Cell adhesion molecules (CAMs) with their adhesive properties and their high functional diversity have been identified as key actors in several of these fundamental processes. Because of their growth‐promoting properties, CAMs play an important role in neuritogenesis. Furthermore, they are necessary to control additional neurite development, regulating dendritic spacing and axon pathfinding. Finally, trans‐synaptic interactions of CAMs ensure cell type‐specific connectivity as a basis for the establishment of circuits processing distinct visual features. Recent discoveries implicating CAMs in novel mechanisms have led to a better general understanding of neural circuit formation, but also revealed an increasing complexity of their function. This review aims at describing the different levels of action for CAMs to shape neural connectivity, with a special focus on the visual system. © 2015 Wiley Periodicals, Inc. The patterning of binocular vision requires distinct molecular pathways for inputs arising from each side of the nervous system. Recent studies have demonstrated important roles for members of the Ten‐m/Odz/teneurin family in the development of ipsilateral retinal projections. Here, we further highlight the significance of this gene family in visual development by identifying a role for Ten‐m4 during the formation of the ipsilateral projection in the mouse. Ten‐m4 was found to be expressed in the retina, dorsal lateral geniculate nucleus (dLGN), superior colliculus (SC), and primary visual cortex (V1) during development. Anterograde and retrograde tracing experiments in Ten‐m4 knockout (KO) mice revealed a specific increase in ipsilateral retinal ganglion cells projecting to dLGN and SC. This increase was most prominent in regions corresponding to temporal retina. Consistent with this, EphB1 expression in the retina around the time of decussation was enhanced in this temporal region for KO mice, suggesting that the increased size of the ipsilateral population arises due to an increased number of retinal ganglion cells remaining ipsilaterally at the optic chiasm due to EphB1‐mediated repulsion. The ectopic ipsilaterally targeted retinal ganglion cell projection observed in Ten‐m4 KOs was associated with changes in response to ethologically relevant visual stimuli. Together, these data demonstrate a requirement for Ten‐m4 in the establishment of ipsilateral projections from the retina, which likely acts in combination with other Ten‐m members (Ten‐m2 and Ten‐m3) to promote the formation of functional binocular circuits.
