Temporal progression of hypothalamic patterning by a dual action of BMP

Abstract: In the developing chick hypothalamus, Shh and BMPs are expressed in a spatially overlapping, but temporally consecutive, manner. Here, we demonstrate how the temporal integration of Shh and BMP signalling leads to the late acquisition of Pax7 expression in hypothalamic progenitor cells. Our studies reveal a requirement for a dual action of BMPs: first, the inhibition of GliA function through Gli3 upregulation; and second, activation of a Smad5-dependent BMP pathway. Previous studies have shown a requirement fo… Show more

“…Therefore, one interpretation may be that BMP4 treatment during the early differentiation stage in our culture prevents hESC-derived forebrain neuroepithelium from adopting the telencephalic fate and redirects it into retinal progenitors. In the early embryo, BMP signals are expressed near the anterior neural ridge and in the prechordal mesoderm, and are implicated in the patterning of rostral diencephalon of the early embryo 65 , including hypothalamic (diencephalic) induction 66,67 .…”

“…The enhanced activation of Smad1 signaling is predicted to participate in neuronal differentiation of p53 -/ -NSCs, as BMPs (the main activator of Smad1/5/8) are known to regulate neurogenesis in cell context, localization, and stage- dependent manners [9,[38][39][40]. To determine the function of BMPs in NSCs, we treated WT and p53 -/ -NSCs with 50 ng/ mL of BMP2, which led to an increase in Smad1/5/8 phosphorylation ( Supplementary Fig.…”

“…Wnt signaling also governs regionalization of the nascent hypothalamus, directing the emergence of posterior (mamillary) hypothalamus throughout development, Shh and multiple different bone morphogenetic proteins (BMPs) show dynamic spatio-temporal profiles. Expression of Shh and Shh signaling components is negatively regulated by BMP signaling (Manning et al, 2006;Ohyama et al, 2008). Studies in the limb bud show that cross talk between the BMP and Shh pathways is critical for integrated growth, pattern, and differentiation (Bénazet et al, 2009), and raises the possibility that a similar cross talk between BMP and Shh signaling regulates proliferation and spatial patterning of hypothalamic neural progenitors in a concerted manner.…”
