Temporal Control of Gene Silencing by in ovo Electroporation

Abstract: The analysis of gene function during embryonic development asks for tight temporal control of gene expression. Classic genetic tools do not allow for this, as the absence of a gene during the early stages of development will preclude its functional analysis during later stages. In contrast, RNAi technology allows one to achieve temporal control of gene silencing especially when used with oviparous animal models. In contrast to mammals, reptiles and birds are easily accessible during embryonic development. We h… Show more

“…To start with, we tested three cationic lipids, DOTAP (Bollérot et al, 2006), DOTMA, and DODAC. (Inoue et al, 2001;Scaal et al, 2004;Baeriswyl et al, 2008;Sauka-Spengler and Barembaum, 2008). A variety of plasmids and nucleic acids can be delivered.…”

“…There is no detailed analysis of avian Dicer specificity and activity, which have to be inferred indirectly from other results. Chicken Dicer can process both, long dsRNA and miRNA precursors, as evidenced by induction of RNAi with long dsRNA (Pekarik et al, 2003;Mauti et al, 2008) and hundreds of avian miRNAs in the miRBase.…”

“…What can be inferred from those studies is that birds have the complete molecular mechanisms for canonical RNAi and can efficiently execute it. This is evidenced by efficient knock-downs with long dsRNA (Pekarik et al, 2003;Mauti et al, 2008).…”
