Temporal and tissue specific gene expression patterns of the zebrafish kinesin-1 heavy chain family, kif5s, during development

Abstract: Homo- and heterodimers of Kif5 proteins form the motor domain of Kinesin-1, a major plus-end directed microtubule motor. Kif5s have been implicated in the intracellular transport of organelles, vesicles, proteins, and RNAs in many cell types. There are three mammalian KIF5s. KIF5A and KIF5C proteins are strictly neural in mouse whereas, KIF5B is ubiquitously expressed. Mouse knockouts indicate crucial roles for KIF5 in development and human mutations in KIF5A lead to the neurodegenerative disease Hereditary Sp… Show more

“…Using in situ hybridization, we detected mRNA expression of this gene from 24 h postfertilization onward in the spinal cord (Fig. 3A), consistent with a recently published expression pattern (Campbell and Marlow 2013). At 3 dpf, kif5aa is broadly expressed in the brain, while BAC transgenesis using the medaka (Oryzias latipes) ortholog showed additional kif5aa transcription in the spinal cord and motoneurons at later stages of development (Kawasaki et al 2012).…”

“…This multicolor labeling strategy can be easily applied to neurobiology studies to induce targeted mutations in single neurons and directly compare loss-of-function and wild-type phenotypes in the same animal. To test this potential application, we targeted the genomic locus coding for the motor protein Kinesin family member 5A, a (kif5aa) (Campbell and Marlow 2013;Auer et al 2015), whose inactivation triggers the reduction of RGC axon arbor complexity via a cell-autonomous mechanism (Auer et al 2015). To target the kif5aa gene with the 2C-Cas9 system in single RGC, we used the Tg(isl2b:gal4) line.…”
