Temperature Significantly Regulates Gene Expressions of Vertebrae Chondrocytes and Osteoblasts in Juvenile Golden Pompano (Trachinotus ovatus)

Abstract: To elucidate the possible molecular reaction of vertebral fusion in juvenile golden pompano at high temperature, we examined the changes in growth, vertebral fusion incidence, histological differences and transcription levels of genes related to bone development in juvenile golden pompano at 27 and 33°C for 30 days, respectively. The growth rate of juvenile fish was faster when the water temperature was 33°C, but the incidence of vertebral fusion was higher. Prolonged high water temperature reduced the osteoge… Show more

“…Nine typical DEGs and b-actin (reference gene) were randomly selected for quantitative real-time PCR (qRT-PCR) to verify the accuracy of the transcriptome and DGEs data, and the primers were designed using the Primer 5 (Table 1). RT-PCR was conducted using an RT-PCR system (Analytik Jena GmbH, Germany) with SYBR Green (Tiangen Biotech Co., Ltd., China), and the reaction conditions were referred to in Han et al (2021). The 2 -DDCT method was used to calculate the relative expression levels of the DEGs.…”

“…Scleral cartilage contains a unique morphology that incorporates multiple elements of cartilage types 34 . In teleost, hyperthermia can increase the occurrence of vertebral fusion and upregulates the expression of chondrogenesis markers including bone morphogenetic proteins (BMP) 4 and 5, Sox9, Basic helix-loop-helix transcription factor (Twist), and Runx family transcription factor (Runx) 2 35 . Thus, further characterization of scleral cartilage could provide valuable insight into teleost ocular biology.…”
