Temozolomide Alone or Combined with Capecitabine for the Treatment of Advanced Pancreatic Neuroendocrine Tumor

Abstract: Background: The combination of capecitabine (CAP) with temozolomide (TEM) chemotherapy in advanced pancreatic neuroendocrine tumors (PanNET) relies on limited evidence. We compared TEM-CAP to TEM alone in patients with advanced PanNET. Methods: Consecutive patients with advanced PanNET treated with TEM or TEM-CAP between 2004 and 2017 in three expert centers were included. Progression-free survival (PFS), tolerance, tumor response, and overall survival were compared between the two groups. Propensity-based ana… Show more

“…Somatostatin analogs, peptide receptor radionuclide and everolimus have been demonstrated to effectively improve the prognosis of pNET [13]. Combined chemotherapy with temozolomide and capecitabine is recently reported to prolong the progression-free survival of pNETs [14,15]. Moreover, our study also shows unmarried patients had signi cantly higher death risks than the married, which coincides with previous reports suggesting that social support could potentially impact cancer survival signi cantly [16].…”

“…TMZ is widely used for the treatment of various tumors, such as adult refractory pleomorphic glioblastoma, anaplastic astrocytoma, pancreatic neuroendocrine neoplasm and invasive pituitary tumor and pituitary cancer (16,20,21). The cytotoxic activities of TMZ lies in DNA methylation and failure of mismatch repair (22,23).…”
