Telomerase-Dependent 3′ G-Strand Overhang Maintenance Facilitates GTBP1-Mediated Telomere Protection from Misplaced Homologous Recombination

Abstract: At the 39-end of telomeres, single-stranded G-overhang telomeric repeats form a stable T-loop. Many studies have focused on the mechanisms that generate and regulate the length of telomere 39 G-strand overhangs, but the roles of G-strand overhang length control in proper T-loop formation and end protection remain unclear. Here, we examined functional relationships between the single-stranded telomere binding protein GTBP1 and G-strand overhang lengths maintained by telomerase in tobacco (Nicotiana tabacum). In… Show more

“…In contrast, sorghum, rice, and maize show non-Rabl patterns. In the case of tobacco, the telomeres are localized at random in the nucleus in a typical non-Rabl pattern (Fujimoto et al 2005, Lee and Kim 2013, Schrumpfová et al 2014. The signal pattern observed with dCas9-FP for telomeres was similar to previous telomere localization patterns in tobacco.…”

“…Furthermore, two‐dimensional (2‐D) pulse‐field gel electrophoresis assays detected no generation of extra‐chromosomal telomeric circles (t‐circles) in both wild type and osrad51d mutant plants (Figure S5). t‐circles result from abnormal inter‐telomeric HR (Zellinger et al ., ; Lee and Kim, , ). osku70 mutant rice exhibit developmental defects in both vegetative and reproductive organs with numerous abnormal anaphase telomeric bridges (Hong et al ., ).…”

“…In addition to the telomerase-mediated telomere elongation, multiple other processes can influence telomere G-overhang length and the terminal sequences 3 . Therefore, telomerase activity is necessary for the normal length of telomere G-overhang in certain eukaryotic organisms 33 , 34 but not others 35 37 . Interestingly, we found that T .…”

“…Indeed, expression of telomerase results in longer G-overhangs at the leading daughter telomeres in human cells 32 while dysfunctional telomerase leads to shorter telomere G-overhangs in tobacco and S . castellii 33 , 34 . However, telomerase deletion does not affect telomere G-overhang length in S .…”
