Technologies to improve the sensitivity of existing chromatographic methods used for bioanalytical studies

Abstract: Chromatographic method has long been recognized as the most widely used separation method in bioanalytical research. However, the relatively low sensitivity of existing chromatographic methods remains a significant challenge, as the requirements for experimental procedures become more demanding. This review discusses the main causes for the low sensitivity of chromatographic methods and aims to introduce different technologies for enhancing their sensitivity in the following aspects: (i) different pretreatment… Show more

“…In fact, there is a great biological variation in these matrices, and the direct recovery of the drug from the tissues is difficult. Thus, the use of skin layer matrices spiked with the analyte is the procedure recommended by the health guidelines and was adopted in this study (Chen, Gao, & Zhong, 2020;FDA, 2018;van Nuland, Rosing, Schellens, & Beijnen, 2020). “…Common detectors used in LC include UV-VIS detectors and diode array detectors (DAD), which use the principle that a physical or chemical property of the sample differs from the mobile phase to achieve the separation of substances [ 75 , 76 ]. This technique has been used to detect polar compounds with high sensitivity, selectivity, and reproducibility [ 77 , 78 ]. The choice of mobile phase and chromatographic column are the main two important parts.…”

“…Derivatization is not always desirable, but sometimes necessary to solve the problem of determining of target analytes from biomatrix (Chen, Gao, & Zhong, 2020). Derivatization may solve some of the problems, but meanwhile it complicates the analytical procedure by adding more operations to the sample preparation process.…”
