Technical note: Comparison of 3 methods for analyzing areas under the curve for glucose and nonesterified fatty acids concentrations following epinephrine challenge in dairy cows

Abstract: The objective of the study was to compare 3 methods for calculating the area under the curve (AUC) for plasma glucose and nonesterified fatty acids (NEFA) after an intravenous epinephrine (EPI) challenge in dairy cows. Cows were assigned to 1 of 6 dietary niacin treatments in a completely randomized 6 × 6 Latin square with an extra period to measure carryover effects. Periods consisted of a 7-d (d 1 to 7) adaptation period followed by a 7-d (d 8 to 14) measurement period. On d 12, cows received an i.v. infusio… Show more

“…Long‐term fasting test, glucose tolerance test (GTT), and insulin tolerance test (ITT) were performed at 9, 10, and 11 weeks, respectively, according to the protocol . The total area under the glucose curve was calculated by using the trapezoidal method . The homeostasis model assessment for insulin resistance (HOMA‐IR) was determined by using the formula: HOMA‐IR = fasting blood glucose levels (mmol/L) × fasting blood insulin levels (mIU/L)/22.5 .…”

“…The area under the curve (AUC) was 3 calculated for both NEFA and BHBA concentrations using the trapezoidal rule as described by Cardoso et al (2011). If an individual cow was missing more than one-third of the variables, data for that cow were eliminated from the data set before analysis.…”

“…Mice were injected with d -glucose (20% in saline, ip) or human insulin (Novolin, 0.0125 µM/g body weight, ip) and blood glucose was quantified at 0–120 min. Total area under the curve (AUC; arbitrary units), which is obtained without the presence of a baseline, calculates the area below the observed concentrations and, in comparison studies, was reported to compare favorably to the positive incremental area method for statistical analyses of glucose tolerance data (Cardoso et al 2011). …”
