TCL1 transgenic mouse model as a tool for the study of therapeutic targets and microenvironment in human B-cell chronic lymphocytic leukemia

Abstract: Chronic lymphocytic leukemia (CLL) is a B-cell malignancy with a mature phenotype. In spite of its relatively indolent nature, no radical cure is as yet available. CLL is not associated with either a unique cytogenetic or a molecular defect, which might have been a potential therapeutic target. Instead, several factors are involved in disease development, such as environmental signals which interact with genetic abnormalities to promote survival, proliferation and an immune surveillance escape. Among these, PI… Show more

“…Recently developed GEMMs use tissue specific approaches to dysregulate gene expression in specific cell types or tissues and provide temporal control of Cre‐recombinase‐mediated excision and disease onset . These endow the study of tumor initiation and provide an intact microenvironment that contains stromal and immune populations more representative of disease propagation in patients . These models endow the study of potential modifier effects resulting from multiple genetic lesions frequently encountered in patients.…”

“…These models have been instrumental in the identification of the mechanisms of leukemogenesis, providing advances in understanding leukemic progression, the role of the niche, and aiding in the identification of novel therapeutic targets. Many of the referenced models have also contributed key insights into stromal remodeling, detailed below .…”

“…To help decipher the pathogenic mechanisms of disease and evaluate the efficacy and mechanisms of action of novel therapies, researchers have developed mouse models that resemble human CLL (12,13). The Eµ-TCL1 transgenic (TCL1-tg) mouse develops a leukemia that is highly similar to the aggressive form of human CLL (14)(15)(16).…”

“…Activation of the PI3K/AKT and MEK/ERK pathways is common in different types of cancer (20)(21)(22)(23) and, in solid tumors, it is often associated with poor prognosis (24). AKT and ERK play crucial roles in cell proliferation, metastasis, angiogenesis, and drug response.…”
