TCF-1 Controls ILC2 and NKp46+RORγt+ Innate Lymphocyte Differentiation and Protection in Intestinal Inflammation

Abstract: Innate lymphocyte populations play a central role in conferring protective immunity at the mucosal frontier. In this study, we demonstrate that T cell factor 1 (TCF-1; encoded by Tcf7), a transcription factor also important for NK and T cell differentiation, is expressed by multiple innate lymphoid cell (ILC) subsets, including GATA3+ nuocytes (ILC2) and NKp46+ ILCs (ILC3), which confer protection against lung and intestinal inflammation. TCF-1 was intrinsically required for the differentiation of both ILC2 an… Show more

“…1). Progressive commitment to the ILC lineage is also dependent, to varying degrees, on a multitude of transcription factors including GATA‐3, TOX, TCF‐1 and NFIL316, 17, 18, 19, 20, 21 (and reviewed in detail in references 10 and 22). …”

“…For example, GATA-binding protein 3 (GATA3) and T cell factor 1 (TCF1) have both been identified as important for regulating ILC2 development (23,24). However, both of these factors have been demonstrated previously to be essential for normal T-cell development, and further investigation has revealed that GATA3 and TCF-1 are required for the generation of a progenitor downstream of the common lymphoid progenitor (CLPs) but upstream of the Id2 1 ILC precursor and are required for multiple ILC subsets (25,26) (Figure 1). …”

“…17 Lymphocytes were isolated from colon lamina propria as previously described. 26 Liver-infiltrating lymphocytes were isolated by centrifugation liver cell suspension on a 33.75% isotonic Percoll (GE Healthcare Biosciences) density gradient for 12 minutes at 693g at room temperature.…”
