Taxonomical re-examination of the genus Neofusicoccum in Japan

Abstract: Neofusicoccum (Botryosphaeriaceae) is a genus of plant pathogenic fungi associated with various woody plants that causes shoot blight, canker, and dieback of fruit and forest trees (Phillips et al., 2008). The genus was established by Crous et al. (2006), based on the type species N. parvum (Pennycook & Samuels) Crous, Slippers & A. J. L. Phillips, and is closely related to the genus Fusicoccum. Neofusicoccum is characterized by fusiform to ellipsoid aseptate conidia and "dichomera-like" synanamorphs, which se… Show more

“…Newly generated in this study were combined using SeqMan v. 7.1.0, and reference sequences (Table 1 ) were downloaded from GenBank, according to the recent publication ( Hattori et al 2020 ; Norphanphoun et al 2020 ; Crous et al 2021 ; Bhunjun et al 2022 ; Nguyen et al 2022 ; Tan and Shivas 2022 ; Zhang et al 2022 ). Alignments were done by MAFFT v. 7.036 ( https://maft.cbrc.jp/alignment/server/ ) using default settings and manually improved using MEGA v.7.0 ( Kumar et al 2016 ).…”

“…Subsequently, Shang (1987) transferred G. laricina to the genus Botryosphaeria based on the morphological characteristics of its sexual state noted by Luttrell (1973). Hattori et al (2021) reexamined the specimens of G. laricina, including the syntype specimens of P. laricina (IUM-FS515, IUM-FS516, IUM-FS517, IUM-FS518, IUM-FS519, and IUM-FS520). In their study, a multi-locus molecular phylogenetic analysis using ITS, rpb2, tef1-α, and tub2 sequences was conducted with newly isolated strains of G. laricina to resolve taxonomic problems associated with this species.…”

“…Targeted sequences of ITS and partial sequences of tef1 , tub2 , and rpb2 regions were amplified. The PCR and sequencing analysis conditions were the same as those used by Hattori, Ando, and Nakashima (2021) . The obtained sequences were further assembled, concatenated, and aligned with 82 sequences from the genus Lasiodiplodia retrieved from the DNA Data Bank of Japan using MEGA v. 5.2 software ( Tamura et al, 2011 ).…”
