Here, we study karyotype divergence in the closely related genera Brasiliorchis, Christensonella and Trigonidium belonging to subtribe Maxillariinae of subfamily Epidendroideae (Orchidaceae). We compare karyotypes in 15 species by (1) measuring 1C genome sizes, (2) mapping the distribution of 4′,6‐diamidino‐2‐phenylindole and chromomycin A3 chromosome bands and (3) localizing 5S and 45S nuclear ribosomal DNA (rDNA) sequences using fluorescent in situ hybridization. Recently, phylogenetic studies have been conducted to resolve species and genera relationships in subtribe Maxillariinae. We used these phylogenetic trees to map the cytogenetic characters in an evolutionary framework. This has enabled a better understanding of the patterns of genomic divergence in the group. Genome sizes range from 1C = 1.85 to 4.1 pg. The largest, B. schunkeana, shows evidence of genome upsizing, probably through the acquisition of tandem repeats that now form large 4′,6‐diamidino‐2‐phenylindole‐positive blocks of heterochromatin. Our cytogenetic data are consistent with a base chromosome number of 2n = 40, although Christensonella is characterized by a dysploid reduction in chromosome number to 2n = 36. The number of 5S and 45S rDNA sites is variable between species, consistent with high rates of karyotype divergence. © 2012 The Linnean Society of London, Botanical Journal of the Linnean Society, 2012, 170, 29–39.

During the taxonomic revision of the genus Macradenia, we observed that a review was needed concerning four names, M. delicatula,M. paraensis, M. regnellii, and M. rubescens, published by the Brazilian botanist João Barbosa Rodrigues. Original illustrations are designated as lectotypes for M. paraensis, Trichopilia multiflora (a synonym of M. paraensis) and M. rubescens. Macradenia rubescens is treated as a synonym of M. surinamensis, the latter lectotypified here using a Wullschlaegel specimen from GOET. A Warming specimen at W is designated as the lectotype of Warmingia eugenii, and W. loefgrenii and M. regnellii, two synonymys of W. eugenii, are lectotypified from specimens at BR and P, respectively. No original material is known for M. delicatula, and, as a result, the precise application of this name is uncertain.

Are chromosome number and genome size associated with habit and environmental niche variables? Insights from the Neotropical orchids

Background and Aims The entangled relationship between chromosome number and genome size with species distribution has been the subject of study for almost a century but remains an open question due to previous ecological and phylogenetic knowledge constraints. To better address this subject, we used the clade Maxillariinae, a widely distributed and karyotypically known orchid group, as a model system to infer such relationships in a robust methodological framework. Methods Based on literature and new data, we gathered the chromosome number and genome size for 93 and 64 species, respectively. We built a phylogenetic hypothesis and assessed the best macroevolutionary model for both genomic traits. Additionally, we gathered ecological data (preferences for bioclimatic variables, elevation, and habit) used as explanatory variables in multivariate phylogenetic models explaining genomic traits. Finally, the impact of polyploidy was estimated by running the analyses with and without polyploids in the sample. Key Results The association between genomic and ecological data varied depending on whether polyploids were considered or not. Without polyploids, chromosome number failed to present consistent associations with ecological variables. With polyploids, there was a tendency to waive epiphytism and colonise new habitats outside humid forests. The genome size showed association with ecological variables: without polyploids, genome upsize was associated with flexible habits, with higher elevation and with drier summers; with polyploids, genome size increase was associated with colonising drier environments. Conclusions The chromosome number and genome size variations, essential but neglected traits in the ecological niche, are shaped in the Maxillariinae by both neutral and adaptive evolution. Both genomic traits are partially correlated to bioclimatic variables and elevation, even when controlling for phylogenetic constraints. While polyploidy was associated with shifts in the environmental niche, the genome size emerges as a central trait to orchid evolution by the association between small genome size with epiphytism, a key innovation to Neotropical orchid diversification.
