Targeting thiamine-dependent enzymes for metabolic therapies in oral squamous cell carcinoma?

Abstract: This is the first study showing increased expression of TDEs in OSCC. Metabolic targeting of TDEs (including TKTs) by antagonistic compounds like oxythiamine or oxybenfothiamine may be a useful strategy to sensitize cancer cells to common OSCC cancer therapies.

“…The underlying mechanism of the relationship between vitamin K intake and CIN risk needs to be further explored. Some studies have found that vitamin B1 has anticancer effects in myeloproliferative neoplasms [62] and oral SCC [63]. Our study results showed that dietary vitamin B1 intake was signi cantly associated with CIN1 risk.…”

“…OSCCs were stratified in a young (<45 years (16)) female (n=7, Table 1) study cohort and older patients (n=46, Table 2) (18). In the young female study cohort three patients (n=3/7) developed OSCC during or shortly after pregnancy.…”

“…Both LDHA overexpression and LDHB overexpression have been associated with poor survival in OSCC indicating that LDH expression is an general unfavorable factor for OSCC. As currently discussed and demonstrated by others tumors with intensively upregulated aerobic and anaerobic metabolic potential could occur due to genetic events that allow a switch on the entire cellular metabolism with a molecular ‘survival of the fittest’ scenario. In part, a nuclear staining pattern was found for LDHA and LDHB expression in IHC as it is well known that LDH resides in both the cytoplasm and nuclei .…”
