Targeting the Enterohepatic Bile Acid Signaling Induces Hepatic Autophagy via a CYP7A1–AKT–mTOR Axis in Mice

Abstract: Background & AimsHepatic cholesterol accumulation and autophagy defects contribute to hepatocyte injury in fatty liver disease. Bile acid synthesis is a major pathway for cholesterol catabolism in the liver. This study aims to understand the molecular link between cholesterol and bile acid metabolism and hepatic autophagy activity.MethodsThe effects of cholesterol and cholesterol 7α-hydroxylase (CYP7A1) expression on autophagy and lysosome function were studied in cell models. The effects and mechanism of disr… Show more

“…Hepatic cholesterol accumulation causes mitochondrial dysfunction, lysosome impairment, and hepatic injury, and has been implicated in the pathogenesis of liver diseases including non-alcoholic steatohepatitis (11)(12)(13)(14)(15)(16)(17)(18)(19). Here we report that mice deficient in SORT1 were protected against cholesterolinduced liver injury.…”

“…For example, elevated levels of cholesterol 7α‐hydroxylase together with decreased bile acids resulting from cholestyramine feeding were recently shown to result in increased levels of hepatic autophagy. This was shown by Wang et al to promote the lipophagy‐mediated hydrolysis of CEs and the transport of lysosomal cholesterol to the ER.…”

“…(46) Increasing bile acid synthesis induces autophagy through the cytochrome P450 7A1-AKT-mammalian target of rapamycin pathway in mice. (47) However, to our knowledge, no previous report has specifically connected autophagy with canalicular bile salt excretion or PFIC2. Autophagy is required for polarization of hepatocytes cultured in a collagen sandwich system.…”
