Targeting the Administration of Ecdysterone in Doping Control Samples

Abstract: PurposeThe phytosteroid ecdysterone was recently reported to enhance performance in sports and may thus be considered as a substance of relevance in anti-doping control. To trace back an administration of ecdysterone from urine samples analytical properties have been investigated to assess its integration into initial testing procedures (ITP) in doping control laboratories. MethodsAnalytical properties of ecdysterone were evaluated using GC-QTOF-MS and LC-QTOF-MS.Its metabolism and elimination in human were st… Show more

“…Such experiments paved the way, directly or indirectly, for use as a “doping substance” by sportsmen or bodybuilders based on the consumption of uncontrolled dietary supplements [ 162 ]. The recent confirmation of these anabolic results [ 163 ] led the authors to propose the inscription of 20E on the list of doping prohibited substances [ 164 ].…”

“…The final example of a Monitoring substance is ECDY 24 which is often labelled as the food supplement “natural anabolic agent”. Several studies have reported it to increase strength and muscle mass, to reduce fatigue and to ease recovery 25,26 . ECDY can be administered from plant extracts, such as spinach.…”

“…UHPLC-MS/MS again proved as the method of choice as it was already useful for a wide range of applications in pharmacology, toxicology, and forensics [ 31 , 32 , 33 ]. Despite first dilute and inject attempts to reduce the time-consuming sample preparation [ 34 , 35 , 36 , 37 , 38 , 39 ], UHPLC-MS/MS analyses still often utilizes extensive sample preparation to separate the molecule of interest from interfering proteins and potential enzymatic degradation processes [ 40 ]. Our method (A) used for quantitation of docetaxel was successfully validated in terms of selectivity, carry-over, lower limit of quantitation (LLOQ), calibration function, accuracy, and precision according to EMA guidelines for bioanalytical method validation.…”
