Targeting of the 22 kDa integral peroxisomal membrane protein

Abstract: Investigating targeting of the 22 kDa peroxisomal membrane protein (Pmp22p) to the peroxisomal membrane we have confined the targeting signal to amino acid residues 16^37 located in the N-terminal cytoplasmic tail. Comparison of Pmp22p orthologous sequences revealed a conserved motif Y3xL3xP3x(KQN) which might represent the core of this targeting signal not found so far in other Pmps. Fusion of the Pmp22p N-terminal tail to the C-terminal portion of Pmp22p which per se is not targeted to peroxisomes, conveys p… Show more

“…Interestingly, the Pex19p-binding site motif resembles a motif of hydrophobic and basic amino acids that has been implicated recently in targeting of mammalian PMPs (Pause et al, 2000;Brosius et al, 2002;Biermanns et al, 2003;Landgraf et al, 2003). This opens the possibility that also in higher eukaryotes Pex19p binds the targeting-specific regions of mPTSs, which however needs to be studied in future work.…”

“…The targeting signals of PMPs (mPTS) that direct and insert PMPs into the peroxisomal membrane have been determined for a number of PMPs of several species. Despite some differences, a picture emerged from these studies of a targeting signal consisting of one or more transmembrane domains in conjunction with a short sequence, which contains either a cluster of basic residues or a mixture of basic and hydrophobic amino acids (Dyer et al, 1996;Baerends et al, 2000b;Pause et al, 2000;Honsho and Fujiki, 2001;Wang et al, 2001;Honsho et al, 2002;Biermanns et al, 2003;Murphy et al, 2003). Although some similarities among these basic sequences have been noted, a clear consensus could not yet be deduced.…”

“…via the endoplasmic reticulum; see below). Thus, although there are no doubts that at least PMP22 and PMP70 are inserted into the peroxisomal membrane directly from the cytosol in liver cells (31,49), we reasoned that no assumptions should be made when studying the biogenesis pathway of a given PMP for the first time. These reservations are justified by recent data showing that some peroxisomal proteins reach the organelle membrane after traveling first through the endoplasmic reticulum (23, 50 -55).…”

“…In addition to the demonstrated importance of matrix-oriented basic clusters in targeting to the peroxisomal membrane, a short cytosol-oriented region adjacent to TMD1 of rat Pmp22 has been shown to contain a peroxisomal targeting signal (14). Similarly, data suggest that a cytosolic region adjacent to the membrane span of Pex15p is required for peroxisomal localization (11).…”

“…Similarly, a basic cluster from the cotton seed ascorbate peroxidase (APX), fused to a synthetic membrane span and carrier protein, targets to punctate and reticular structures, although peroxisomal localization was not demonstrated (13). In contrast, a cytosol-oriented region of 22 amino acids in rat Pmp22 can target a different fragment of the protein, which by itself is not sufficient for targeting, to the peroxisomal membrane (14). Because some peroxisomal membrane proteins integrate into peroxisomal precursor organelles before arriving at peroxisomes (15), a more complex targeting signal may be required to accommodate such a pathway.…”
