Targeting of plasmid DNA‐lipoplexes to cells with molecules anchored via a metal chelator lipid

Abstract: The results show that DODAP-containing lipoplexes incorporating NTA(3)-DTDA enable the engraftment of targeting molecules and the effective targeting of pDNA to cells in serum-containing media, resulting in efficient transgene expression. The strategy may provide a convenient approach for targeting pDNA to cells in vivo in therapeutic applications.

“…Binding assay was carried out to determine the peptide (six histidine-TAT peptide) to DOGS-NTA-Ni ratio as per reported method [24]. In brief, for the binding assay, adherent H460 cells were seeded at a density of 1×10 5 cells/well in a 24-well plate.…”

“…The fluorescent lipid Oregon Green-488-(1,2-dihexadecanoyl-sn-glycero-3-phosphoethanolamine) (OG 488 -DHPE) was purchased from Invitrogen (Eugene, OR). 1,2-palmitoyl-oleoyl-snglycero-3-phosphatidylethanolamine was reacted with Alexa Fluor-647 carboxylic acid succinimidyl ester (Invitrogen) and purified to produce Alexa Fluor-647-1,2-palmitoyl-oleoyl-sn-glycero-3-phosphatidylethanolamine (AF 647 -DOPE), as previously described (30). Paraformaldehyde was from BDH Chemicals (Kilsyth, Victoria, Australia).…”

“…Other promising innovative approaches for ligating ligands to the liposome surface include transpeptidation catalyzed by sortase A [79] and the chelating strategy employing His-tagged proteins and nitrilotriacetic acid (NTA 3 ) modified liposomes [8486]. The latter strategy has opened the opportunity to modify liposomes with a vast variety of His-tag possessing proteins (or peptides).…”
