Targeting Lysosomes to Reverse Hydroquinone-Induced Autophagy Defects and Oxidative Damage in Human Retinal Pigment Epithelial Cells

Abstract: In age-related macular degeneration (AMD), hydroquinone (HQ)-induced oxidative damage in retinal pigment epithelium (RPE) is believed to be an early event contributing to dysregulation of inflammatory cytokines and vascular endothelial growth factor (VEGF) homeostasis. However, the roles of antioxidant mechanisms, such as autophagy and the ubiquitin-proteasome system, in modulating HQ-induced oxidative damage in RPE is not well-understood. This study utilized an in-vitro AMD model involving the incubation of h… Show more

“…Actually, the effect of dB on autophagosome accumulation in MdA-MB-231 and McF7 cells was relatively similar, based on the results of western blot analysis. Various studies have revealed that alkalization of lysosomes, defects in lysosomal proteolytic activity, and delayed trafficking of autophagosomes to lysosomes easily lead to the limitation of autophagosomes-lysosome fusion (48,49). However, the data from the AO and LysoTracker Red staining assays in the present study demonstrated that the pH was not altered in response to dB treatment.…”

“…Interestingly, the expression levels of PINK1 protein levels were only increased in manifest HD. Protein LAMP2 is essential in controlling autolysosome formation and degradation of autophagosomes (Abokyi et al, 2021). The levels of LAMP2 were decreased in manifest HD and control cells after treatment with the autophagy inhibitor CQ indicating downregulation of the lysosomal membrane-stabilizing and loss of lysosomal membrane integrity and function at a greater level than in pHD (Figure 2b).…”

“…In vitro or cell-based models (ARPE19) may be useful for determining the effect of hydroxychloroquine exposure on cellular omics (gene expression) in the short term (27). These models may help elucidate the pathways to toxicity.…”
