Targeting IL-17 attenuates hypoxia-induced pulmonary hypertension through downregulation of β-catenin

Abstract: BackgroundThe role of interleukin 17 (IL-17) in hypoxic pulmonary hypertension (HPH) remains unclear. This study is designed to explore whether IL-17 is a potential target for HPH treatment.MethodsClinic samples from the lung tissue and serum were obtained from qualified patients. Western blotting, immunohistochemistry and/or ELISA were used to measure the expression of relevant proteins. HPH models were established in C57BL/6 wild-type (WT) and IL-17 −/− mice and were treated with exogenous recombinant mouse … Show more

“…HPAH and CTEPH are two common types of PH (1). PASMCs in HPAH and CTEPH exhibit proliferation and migration phenotypes similar to those of cancer cells (25,26). In the present study, EZH2 expression levels were increased in both hypoxia-induced and patient-derived PASMCs, which is consistent with a previous study in a hypoxia-induced PAH mouse model (11).…”

“…Th17 cytokines are featured as IL-17 and GM-CSF. Th17 cells infiltration and IL-17 elevation are observed in the clinic and experimental PAH/PH ( 36 , 37 ). Over-expressing IL-17 or blocking IL-17 in mice regulates the PH development through directly affected PAECs and PASMCs function ( 37 ).…”

“…Consistent with published data, the enrichment of these DEGs in GO terms, such as leukocyte migration (20,21), collagencontaining ECM (22,23), cytokine activity (24,25), and antioxidant activity (26), confirms their involvement in the development of IPAH. Besides, enrichment of the identified DEGs in some KEGG pathways, such as the chemokine signaling pathway (24,25), IL-17 signaling pathway (27,28), and NFkappa B signaling pathway (29,30), also suggests the relevance in IPAH pathogenesis. Based on GO and KEGG analysis results, we suggest that these DEGs are closely associated with immune response and IPAH development.…”
