Targeting head and neck tumoral stem cells: From biological aspects to therapeutic perspectives

Abstract: Head and neck squamous cell cancer (HNSCC) is the sixth most common cancer in the world. Effective therapeutic modalities such as surgery, radiation, chemotherapy and combinations of each are used in the management of the disease. In most cases, treatment fails to obtain total cancer cure. In recent years, it appears that one of the key determinants of treatment failure may be the presence of cancer stem cells (CSCs) that escape currently available therapies. CSCs form a small portion of the total tumor burden… Show more

“…Once the disease has relapsed or becomes metastatic, patients' median survival time is about 9 months [3], and there are few treatment options. While therapeutic strategies such as surgery, radiation and chemotherapy when applied alone or in combination are useful for managing the disease [4], in most cases, they fail to obtain a cure. The most commonly used treatments for head and neck cancers are platinum agents, taxanes, 5-fluorouracil, methotrexate and cetuximab.…”

“…Według danych polskich z roku 2014 nowotwory GiS stanowią około 9% wszystkich nowotworów złośliwych w populacji mężczyzn oraz około 5% w populacji kobiet. Wznowy choroby obejmują blisko 60% chorych, a rozwój przerzutów występuje u 20% (1,2). Są to guzy pochodzące z nabłonka jamy ustnej, gardła, języka i krtani.…”

“…HNSCC is the sixth most common cancer in the world and remains a major health problem globally with an estimated 500,000 new cases diagnosed yearly. 1,2 Although effective therapeutic modalities such as surgery, radiation, chemotherapy, and therapeutic combinations of these approaches are used in the management of this disease, recurrent HNSCC is almost inevitable and becomes resistant to further therapies. Systemic chemotherapy, which was considered an adjuvant in the past, is currently the major approach for organ preservation strategies for HNSCC patients; it is often combined with definitive radiotherapy or induction treatments, and is no longer reserved for palliative purposes.…”
