Targeted Repair of Vascular Injury by Adipose‐Derived Stem Cells Modified with P‐Selectin Binding Peptide

Abstract: Percutaneous coronary intervention for coronary artery disease treatment often results in pathological vascular injury, characterized by P‐selectin overexpression. Adipose‐derived stem cells (ADSCs) therapeutic efficacy remains elusive due to poor ADSCs targeting and retention in injured vessels. Here, conjugated P‐selectin binding peptide (PBP) to polyethylene glycol‐conjugated phospholipid derivative (DMPE‐PEG) linkers (DMPE‐PEG‐PBP; DPP) are used to facilitate the modification of PBP onto ADSCs cell surface… Show more

“…Primary fibroblasts and ADSCs were successfully isolated from human tissues and identified separately. ADSCs were identified by CD73+, CD90+, CD105+, CD31−, CD34− and CD45− [ 44 , 45 ], and by their ability to differentiate into adipogenic, chondrogenic and osteogenic ( Supplementary Fig. 1 ).…”

“…The latest decade has witnessed rapid progress in biomaterials and medical devices, in both fundamental research [ 44–51 ] and applications [ 52–59 ]. While medical materials have been much developed [ 60–68 ], it is still challenging to find a biomaterial-related cardiovascular device appropriate for interventional treatment.…”

“…Cells were incubated in a humidified hypoxia incubator (Thermo Scientific) with 5% CO 2 , 1% O 2 , and 94% N 2 at 37 °C for 6 h and then reoxygenated with 95% air and 5% CO 2 for an additional 12 h, as previously reported. [ 20 ]…”
