Targeted Proteomic Analyses of Nasal Lavage Fluid in Persulfate-Challenged Hairdressers with Bleaching Powder-Associated Rhinitis

Abstract: Hairdressers have an increased risk for developing airway symptoms, for example, asthma and rhinitis. Persulfates, which are oxidizing agents in bleaching powder, are considered important causal agents for these symptoms. However, the underlying mechanisms are unclear. The aim was therefore to measure proteomic changes in nasal lavage fluid from persulfate-challenged subjects to identify proteins potentially involved in the pathogenesis of bleaching powder-associated rhinitis or candidate effect biomarkers for… Show more

“…GSTP1 gene activation is mainly regulated by Nrf2, and the functional association between GSTP and Nrf2 proteins supports the regulatory role of GSTP in the adaption response to cellular stresses produced in the course of inflammatory and oxidative reactions . As in the present study, GSTP was decreased in NLF after the persulphate challenge …”

“…Few studies have reported the proteome of work‐related airway diseases. Mörtstedt et al performed targeted proteomic analyses of NLF before and after a persulfate challenge among hairdressers with bleaching powder‐associated rhinitis as compared to healthy hairdressers and atopic subjects. They detected changes in proteins related to inflammatory responses and oxidative stress, but the changes were not specific to rhinitis patients.…”

“…The p -value list provided by the ibb test was subsequently subjected to a multiple testing adjustment based on a sequential goodness of fit (SGoF) metatest (Carvajal-Rodriguez et al, 2009) using the SGoF+ software (v.3.8) with default parameters (Carvajal-Rodriguez and de Una-Alvarez, 2011). This metatest has been successfully applied to large RNA-seq and proteomic datasets (Shi et al, 2012; Mortstedt et al, 2015), and was applied in this study in view of its suitability for small sample size and ability to maintain a high statistical power when increasing the number of test. An adjusted p -value < 0.05 was considered as the threshold for statistical significance of differential results.…”

“…Nasal samples, including tissues,6 lavages,7 swabs8 and suctions,910 have been used to identify the CRS endotypes. However, the invasive procedures ( e.g., biopsy) are associated with the risk of infection and must be performed by experts 11.…”

“…Nasal lavages, swabs and suctions have alternatively been obtained; these are constantly available, and sampling does not involve a risk of infection 11. Moreover, the proteins in nasal samples like the antimicrobial proteins1213 and immunoglobulins14 could reflect the upper airway diseases 7891015. Thus, the nasal secretions are valuable when studying CRS endotypes 11.…”
